hdc decarboxylates histidine through use of plp cofactor bound in schiff base lysine 305. histidine initiates reaction displacing lysine 305 , forming aldimine plp. histidine s carboxyl group leaves, forming carbon dioxide. finally, plp re-forms original schiff base @ lysine 305, , histamine released. mechanism similar employed other pyridoxal-dependent decarboxylases. in particular, aldimine intermediate common feature of known plp-dependent decarboxylases. hdc highly specific histidine substrate.
